Greene County, like most other counties in Southwest Missouri, is a Republican stronghold. Greene County was won by George W. Bush in 2000 and 2004, almost by two-to-one margins. In 2008, Greene County, like other Southwest Missouri counties, favored John McCain over Barack Obama. Donald Trump won Greene County in the 2016 presidential election by a margin between 60% and 33%. Lyndon B. Johnson, the last Democratic presidential nominee, won Greene County in 1964. The 2004 Missouri constitutional amendment that defined marriage as the union of a man or woman was approved by the Missourians. It passed Greene County with 72.04% of the vote. With 71 percent support from the voters, the initiative was passed and Missouri became the first state that bans same-sex marriage.
Missourians voted in 2006 on a constitutional amendment that would legalize embryonic stem-cell research. It was narrowly defeated in Greene County by 51.62 percent of those who voted against it. With 51 percent support from the voters, the initiative passed narrowly. Missouri became the first state in the country to approve embryonic stem cell research. Greene County has a long history of supporting conservative platforms but its voters have a tendency to support populist causes such as increasing the minimum wage. Missourians voted in 2006 on Proposition B to raise the minimum wage to $6.50 per hour. It passed Greene County with 74.31% of the vote. With 78.99% of the vote in support, the Missouri minimum wage was raised to $6.50 per hour. This proposition passed in every county. Five other states also voted in favor of raising the minimum wage during the same election. In 2020 Greene County was among eight Missouri counties and the only one other than St. Louis, Kansas City, and Columbia that voted yes to Amendment 2 to expand Medicaid. It passed Greene County with 52.3% of the vote, and statewide, with 53.3 percent.
