Greenville was established for the first time in 1819. Although it was originally called Buttsville in the early days, the county seat was established in 1822. The name Greenville was adopted in honor of the South Carolina area where many of the first settlers settled. Fort Dale was the first county seat. It was named after Sam Dale who defended the area during Creek War. Near the Fort Dale Cemetery is the site of Fort Dale, which can be found on the northern side of the city.
Captain William Butler, the county's namesake, was killed in the Creek War. He is buried at the Pioneer Cemetery, just across from the First United Methodist Church in Greenville, the oldest church of Butler County.
Greenville was the home of a satellite camp that held German prisoners during World War II.
